Tube swap question

Hi all , I have a Mesa Express 5:25 + I plugged in yesterday and I noticed the clean mode in channel 1 has lost a lot of volume and crackles a bit when I pick hard.
I am not real knowledgeable about tube amps, but assume it's a bad pre-amp tube as the amp sounds fine in the other channel and modes. I have some extra 12ax7's from a complete tube swap from a classic 30. My question is can I swap out V1 and V2 one at a time with the standby switch on and check which tube is bad without turning the amp on and off or should the amp be turned off completely when pulling tubes.
Don't mean to be dumb, just don't know better and don't want to do something stupid. Any help is most appreciated. Tnx
 
turn the amp off completely, let the tubes cool down, do the swap, turn the amp on...repeat as needed :)
